Do it during off hours.
If you are using CONSISTENT=Y option for export or using commit after importing the whole table then make sure that you create a large rollback segment and make this the only online rollback segment.
Use direct path option to speed up export.

I have previously cloned the database into a test instance. Can I just import the dump file without clearing out any of the old data? - ie will it overwrite the old data with the content of the import file.
